Purifikasi Antigen Outer Membrane Protein (OMP) Dari Isolat Salmonella enterica serovar Typhi
Abstrak
Penelitian ini bertujuan mendapatkan protein murni dari antigen OMP isolat Salmonella enterica serovar typhi (S. typhi) dengan metode fraksinasi amonium sulfat dan dialisis. Fraksinasi dilakukan dengan variasi konsentrasi ammonium sulfat 10–20%; 20-40%; 40–60%, 60–80%, dan 80–100%. Kemudian filtrat dari setiap konsentrasi fraksinasi tersebut dilanjutkan dengan metode dialisis menggunakan membran selofan, kemudian diukur kadar proteinnya dengan metode Lowry. Hasil penelitian menunjukkan bahwa Kadar protein OMP tertinggi diperoleh sebesar 1, 641 mg/ml yaitu pada konsentrasi 20-40%.
