UJI AKTIVITAS ANTIMUTAGENIK EKSTRAK ETANOL DAUN KEMANGI (OCIMUM BASILICUM L.) TERHADAP MENCIT DENGAN MENGGUNAKAN METODE MIKRONUKLEUS ASSAY
Abstract
Penelitian ini bertujuan untuk mengetahui potensi antimutagenik ekstrak etanol daun kemangi terhadap mencit (Mus musculus L.) dengan menggunakan metode uji mikronukleus. Serbuk daun kemangi dimaserasi dengan pelarut etanol. Ekstrak etanol daun kemangi selanjutnya dibuat dalam bentuk suspensi dengan dosis 50 mg/kgBB, 100mg/kgBB dan 200 mg/kgBB. Uji antimutagenik dilakukan dengan metode uji mikronukleus (MN) dengan melihat penurunan jumlah mikronukleus pada 200 sel eritrosit polikromatik (PCE) sumsum tulang paha mencit yang telah diinduksi larutan siklofosfamid 50 mg/kgBB.
Hasil penelitian menunjukkan bahwa pemberian ekstrak etanol daun kemangi
(Ocimum basilicum L.) dengan dosis 50 mg/kgBB, 100mg/kgBB, dan 200
mg/kgBB yang diberikan selama 7 hari secara oral mampu menurunkan jumlah
persentase sel eritrosit polikromatik (PCE) yang bermikronukleus yang diamati
melalui preparat apusan sumsum tulang paha mencit. Ekstrak etanol daun
kemangi (Ocimum basilicum L.) dengan dosis 200 mg/kgBB (7,33 ± 2,88)
menurunkan jumlah sel mikronukleus lebih banyak dibandingkan dengan dosis
50 mg/kgBB (43 ± 19,95) dan 100 mg/kgBB (26,33 ± 3,21).
